How the LightCycler Platform Works

The LightCycler is a plate-based real-time polymerase chain reaction (PCR) amplification and detection platform which monitors the amplification of a targeted DNA molecule during the PCR. Because monitoring is done in real-time, and not at the end as in conventional PCR, LightCycler can be used to accurately evaluate gene quantification and genetic variation analysis with a high rate of reliability.

Our LightCycler 480 platform enables high-performance at medium- to high-throughput with 96- or 384-well plates. It boasts a special arrangement of optical components to ensure the uniform collection of signals across the plate, as well as an innovative PCR thermal block cycler design to precisely controls temperature, and makes analysis independent of the sample position on the plate.

As a real-time PCR platform, LightCycler provides superior methods for gene detection, gene expression analysis, genetic variation analysis, and array data validation.

BioAgilytix uses the Lightcycler platform to rapidly and accurately perform assays for gene quantification, specific to gene detection, absolute quantification, and relative quantification / gene expression analysis. We also leverage it for genetic variation analysis, as the platform allows for melt-curve genotyping, endpoint genotyping, high-resolution melting (HRM), and multiple plate analysis.

The primary benefit of the LightCycler is its ability to quickly generate highly reproducible results for mono- or multicolor applications, as well as multiplex protocols. Its accuracy is supported by cutting-edge thermal block and optical technology, which enables excellent well-to-well homogeneity and maximizes detection sensitivity.

This innovative PCR thermal block cycler design precisely controls temperature to yield exceptional data uniformity, independent of assay formats or real-time PCR applications. The LightCycler also boasts a high-performance optical system that captures all data simultaneously for a whole plate—without the need to scan—to minimize inconsistencies and mechanical instability.

LightCycler uses fluorescence excitation by a high-intensity xenon lamp, paired with a specially developed signal-collection system, to provide maximal sensitivity for any assay format in all channels. It also eliminates the “edge effect” found in most block-based themal-cycling systems, helping to mitigate uncontrolled variables so that the data within and between assays is consistent and reproducible.
