Biological or Clinical Significance:

The proline-rich Akt substrate of 40 kDa (PRAS40) acts at the intersection of the Akt- and mammalian target of rapamycin (mTOR)-mediated signaling pathways.PRAS40 is an insulin-regulated inhibitor of the mTORC1 protein kinase. PRAS40 has been shown to regulate the activity of mammalian target of rapamycin complex 1 (mTORC1), which is a signaling hub connecting insulin signaling and autophagic signaling pathway. PRAS40 has received increasing attention as a potential therapeutic target for the treatment of insulin resistance as it has been demonstrated to reduce systemic hyperglycemia and ameliorate insulin sensitivity of obese mice.
